Overview
Delta Sleep Inducing Peptide (DSIP) is a small naturally occurring nonapeptide (sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu) first isolated in the 1970s from the blood of sleeping rabbits. It acts as a modulator influencing slow-wave sleep, neurotransmitter levels, circadian rhythm, hormone release (including growth hormone), and stress responses. It does not have one clear receptor of its own. Despite five decades of study it has no regulatory approval and the human evidence for it as a sleep aid is weak and inconsistent.
Primary Uses
Sleep promotion
Stress reduction
Hormone regulation
Delta (slow-wave) sleep promotion
Circadian rhythm normalization
Pain modulation
Sleep normalization — promotes delta-wave (deep) sleep and normalizes sleep architecture in subjects with disrupted sleep patterns
Stress modulation — reduces stress-induced cortisol elevation and normalizes ACTH levels
Pain modulation — interacts with opioid systems, showing analgesic effects in some studies
Withdrawal support — explored for alcohol and opioid withdrawal symptom management
