Overview

Exenatide is a synthetic 39-amino-acid peptide (copy of natural exendin-4) originally isolated from Gila monster saliva. It was the first GLP-1 receptor agonist approved for market, sold as Byetta (twice daily) and Bydureon (once weekly). It binds and activates the GLP-1 receptor, triggering glucose-dependent insulin secretion, suppressing excess glucagon, slowing gastric emptying, and increasing satiety. Unlike native GLP-1, exendin-4 resists DPP-4 enzyme degradation, giving it a half-life of roughly 2.4 hours.
