Retatrutide is a once-weekly injectable peptide that acts as a triple agonist targeting GIP, GLP-1, and glucagon receptors. It combines three hormone signals into one molecule to curb appetite, support insulin response, and increase energy expenditure and fat burning. In phase 2 and phase 3 trials, it has produced some of the largest weight loss ever recorded for a drug, up to 28.3% mean weight loss at 80 weeks. It is investigational, made by Eli Lilly, and not yet FDA approved.
