Overview

SS-31 (Elamipretide, Bendavia, MTP-131, FORZINITY) is a cell-permeable, mitochondria-targeted peptide that concentrates in the inner mitochondrial membrane and binds tightly to cardiolipin. By stabilizing cardiolipin, it preserves cristae structure, reduces electron leakage that produces reactive oxygen species, and supports more efficient ATP production. It may also modulate membrane surface electrostatics and limit opening of the mitochondrial permeability transition pore. FDA accelerated approval granted September 2025 for Barth syndrome as the first mitochondria-targeted therapeutic.
